Executive Summary : | This application purpose to test state-of-the-art dual NK cells activation methods through high-throughput-screening-approach (HTS) to reprogramme their effector function to inhibit tumor growth to develop a state-of-the-art future generation immunotherapeutic anticancer regime. This application is prepared during various meeting (in total 13) held between Prof Acharya International collaborates from Russia, Australia and Slovakia. NK cells plays a major role in immune surveillance to effectively remove altered cells which may have potential to develop into tumor, however tumor microenvironment modulates NK cells in the favours of tumor growth. The NK cells will be artificially activated by various cytokines combination to select three best hit combinations (HTS-1, Objective 1), these activated NK cells will be further reprogrammed and sensitized for tumor microenvironment through various nucleoside analogues (HTS-II, Objective 2). Further, the best-hit combination will be proceeded to study the mechanism in vitro, target recognition through RNA seq analysis, complemented by Bio-informatics-cum-docking analysis (Objective 3); and preclinical evidence will be generated in T-cell Lymphoma bearing mice model (objective 4), for supportive evidence to translate the findings in phase-1 clinical trial after completion of this project. |
