Executive Summary : | The study aims to investigate the regulation of PWWP2A in glioblastoma cells, focusing on its role in DNA methylation. Despite the existence of nearly 20 PWWP domain-containing proteins in humans, the regulation of PWWP2A is not well studied at the transcript and translational levels in normal and cancer cells. PWWP2A interacts with HDAC1/2 and MTA1/2, regulates gene expression via histone deacetylation, and binds selectively to H2A.Z-containing nucleosomes with a strong preference for promoters of highly transcribed genes. Preliminary data showed elevated PWWP2A and DNMT3B levels in glioblastoma cells, indicating their oncogenic function. The study also identified two CRE motifs in the promoter region of PWWP2A, suggesting that CREB could also regulate PWWP2A transcription. Additionally, miR-150 downregulates epigenetic regulators, including DNMT3A/B. The proposed study will map the minimal promoter region for PWWP2A transcription, identify transcription factors responsible for PWWP2A transcriptional activation, validate predicted miRNAs mediated downregulation of PWWP2A, and identify potential E3 ubiquitin ligase for PWWP2A protein degradation. |
