Executive Summary : | Hematopoietic differentiation and immune regulation are influenced by transcription factors, epigenetic factors (EFs), and noncoding RNAs. The role of auxiliary subunits of the chromatin remodelling complex, such as the human SWI/SNF complex, is poorly understood. The SMARCD family, which includes three isoforms, is crucial for various biological functions, including development, cell cycle, and immune regulation. SMARCD1 is a negative regulator in myeloid differentiation and leukemia, and its role in myelopoiesis has been previously reported. The proposed study aims to understand the epigenetic mechanisms by which SMARCD isoforms regulate myeloid innate immune responses. The study will use both in vitro and ex vivo systems to knock down SMARCD1 using RNAi and check the expression of pro/anti-inflammatory cytokines. A multi-omics approach will be used to delineate global chromatin and transcription regulation. Co-IPs and mass-spectroscopy will be used to identify co-factors associated with SMARCD1 containing the SWI/SNF complex. The study will identify critical epigenetic switches in innate immune regulation and its physiology, potentially triggering abnormal hematopoiesis and its immunological profile. The focus will also be on SMARCD2 and SMARCD3 to understand their roles in immune regulation and if they have redundant or unique functions. This comprehensive study is the first to understand the role of SMARCD isoforms in myeloid physiology and could pave the way for understanding SMARCD as a potential drug candidate. |
