Executive Summary :

Researchers are studying Fanconi anemia (FA), a rare genetic disorder linked to cancer, to understand the mechanisms of genomic instability. FA proteins repair DNA inter-crosslink damage through a unique pathway. Their previous project proposed a novel s-phase checkpoint pathway involving Fanconi anemia protein G. They found that mitomycin C-mediated ICL damage causes the ATR-mediated phosphorylation of serine 7 of FANCG, which is crucial for its interaction with MCM3AP. The researchers hypothesize that acetylation of MCM3 causes the dissociation of MCM3 from the chromatin, leading to the replication halt or s phase checkpoint.
