Executive Summary :

The present research outlines studies that aim to monitor comprehensively the changes in the flight muscle both at daily and seasonal levels in meeting the energy homeostasis using a latitudinal (Palearctic-Indian) obligate migratory buntings (Emberiza sp.). These songbird migrants undertake a highly energy demanding endurance flight, involving an intense activity of the functioning of flight muscles, which thus are the major site for energy utilization. The research will involve laboratory studies in captive birds maintained under controlled photoperiod and temperature conditions. Birds will be exposed to appropriate photoperiods to reproduce the winter non-migratory and spring migratory life history states (LHss). Continuous behavioural and physiological assays will establish the seasonal LHs, and gene (including RNA-seq and qPCR) and protein (immunocytochemistry, LC-Ms) assays will identify core regulatory molecules and understand daily and seasonal changes to support the intense nocturnal migratory flight. In addition to new data on mechanisms underlying the migratory physiology which hitherto remains unclear, the proposed research will provide molecular entry points to understand the etiology of metabolic disorders in vertebrates, and perhaps in humans.
