Executive Summary :

According to Ayurveda natural products are outstanding, effective, and safe for medicinal use. Bioactive compounds of natural product have big impact on weight loss and obesity. From several thousand years mushrooms have been documented for the nutritional and medicinal values. They are known to have many health benefits like immune supporter, anti-inflammation, helps in manage blood sugar, supports brain health and cognition. Mushroom contains biologically active primary and secondary metabolites which protect human from lifestyle diseases. The crude extract of Inonotus sp. has been shown to have numerous biological activities like anti-hyperglycemic, anti-viral, anti-fungal, anti-tumor, and anti-oxidant. Therefore, it is a good candidate for pharmaceutical or nutraceutical applications. The present study aims to understand the molecular mechanism of exudate of Inonotus spp. on obesity. Human Mesenchymal stem cells will be cultured and differentiated into adipocytes in presence and absence of Inonotus exudate, adipocytic differentiation will determined by oil red O staining and Triglyceride estimation. Further, these samples will be processed for transcriptomic analysis and differentially expressed genes will be determined to understand the detailed molecular mechanism. Finally, this exudate's anti-adipogenic property will be confirmed in mice model. Also, active component of the exudate will be determined which is anti-adipogenic in nature.
