Executive Summary : | Epithelial ovarian cancer (EOC) is the leading cause of mortality in women due to gynecological cancer, with high-grade serous ovarian cancer (HGsOC) accounting for 70-80% of patients. Cancer relapse after primary chemotherapeutic treatment is higher in these cancers, partly due to the presence of Cancer stem Cells (CsCs). Targeting CsCs is an alternative approach to improve patient outcomes. Recent studies have identified CD24 and EPHA1 as cell surface markers for CsCs in HGsOC, and validated them in vitro and in vivo. The precise molecular mechanism and downstream signaling cues of EPHA1 are unexplored. The researchers evaluated the expression of EPHA1 using different ovarian cancer cell lines and found a clear association with CsCs with its marker ALDH1A1. They cloned the extra-cellular EPHA1 domains and tested their constructs in 293 T cells. The researchers plan to use CRIsPR-Cas9 gain of/loss of function screening to define the mechanism and shortlist actional targets for CsC survival in EPHA1 expressing cells and test the efficiency of inhibiting EPHA1 via antibody therapeutic approach. |
