Executive Summary : | The study aims to understand the mechanisms and causality between periodontitis and cardiovascular diseases (CVD). It focuses on the permeability of the transendothelial barrier under the influence of unmodified LDL, the molecular mechanisms involved in P. Gingivalis-induced cardiovascular dysfunction, the roles of endogenous EETs in mitigating Pg-induced cardiovascular dysfunction, and the therapeutic effects of LNPG on Pg-driven cardiovascular dysfunction. The hypothesis is that EG damage has an etiological role in periodontitis-induced cardiovascular diseases and has therapeutic implications. The proposed studies focus on the causative relationship between periodontitis and CVD, preventive strategies of endogenous EETs, and therapeutic interventions for exogenous administration of LNPG. The proposed studies have the potential to expose mechanistic links between the loss of EG integrity and periodontitis-induced cardiac dysfunction and clarify a protective mechanism of EET-mediated stabilization of EG. This project is the first to use LNPG to restore EG to alleviate endothelial dysfunction driven by periodontitis. The study may provide new therapeutic opportunities by capturing the treatment of the glycocalyx of the endothelium, which controls vascular permeability, leukocyte trafficking, and vasomotion. The method presented in this study has the potential to completely change the way glycocalyx restoration and treatments are approached. |
