Executive Summary :

Loose smut (Ls) is a significant wheat disease with increasing incidence and direct effect on yield. Breeding for Ls resistance has been given low priority due to effective chemical control, but developing resistant cultivars is the most desirable and environmentally friendly strategy. Genetic analysis of potential donors remains inadequate due to tedious population development requiring two seasons. Marker Assisted selection (MAs) should be applied provided tagged genes are available. In durum wheat, very few known genes are available in Ls internationally, and all reported genes have been tagged from durum wheat only. A study using an Infiniumi select 90K single nucleotide polymorphism (sNP) and composite interval mapping analysis revealed three significant quantitative trait loci (QTL) for loose smut resistance on chromosomes 3A, 6B, and 7A. This is the first attempt in durum wheat to identify and map loose smut resistance QTL using a high-density genetic map. This project aims to identify genes from four mapping populations (NILs) that are well phenotypically characterized over the last 10 years. The identified linked sNPs will be validated on NIL populations and used to develop MAs-friendly KAsP markers. The readiness level of the study includes well phenotyped and genetically characterized NILs available just a step away from known gene(s). The tagged genes -NILs with sNP as markers will be shared across wheat breeders for incorporation in the individual wheat varietal pool. The QTLs can be used to transfer loose smut resistance to commercial cultivars, making them more environmentally friendly and beneficial for farmers across the country.
