Executive Summary :

In this proposal, we aim to develop a multifunctional theranostic platform utilizing polyion vesicles loaded with gold (Au) semishells. This unique approach exploits the strong plasmonic scattering of the gold nanoparticles (GNPs) in the near-infrared (NIR) region, caused by localized surface plasmon resonance (LsPR). This feature offers potential for improved cancer imaging, diagnostics, and treatment. The polyion vesicles encapsulate a photosensitizer within an inner aqueous phase, optimizing them for use as nanocontainers with adjustable permeability and enabling photo-induced release of photoactive agents within cellular systems to increase therapeutic efficacy. In addition, we aim to align the photo-properties of both the Au semishells and the photosensitizer for simultaneous activation via single laser irradiation. For effective treatment, photodynamic therapy (PDT) leverages reactive oxygen species produced when photosensitizers react with tissue oxygen upon light exposure, while photothermal therapy (PTT) relies on local temperature increase. We anticipate that both strategies will produce positive therapeutic results. With efficient photosensitizer loading, increased cellular uptake, simultaneous image acquisition (NIR fluorescence, thermal, photoacoustic), and a synergistic PTT or PDT approach, our proposed platform offers potential advancements in phototherapy applications.
