Executive Summary : | The project aims to develop a safe media additive using Azadirachta indica neem, Terminalia arjuna arjun, Ocimum sanctum tulsi, and Aegle marmelos bael leaf extract for optimal laboratory production of bovine embryos. The project will screen medicinal plants for green-synthesis of metal nanoparticles, extract active compounds, immobilize nanoparticles onto biodegradable polysaccharides, and conduct qualitative and quantitative characterization using various techniques. The study will also examine the effects of biogenic nanocomposites on in vitro maturation, sperm capacitation, and fertilization of bovine oocytes, focusing on embryo production and quality.
The study aims to assess the toxicity of nanoparticles and nanocarriers in biological systems, using various invivo and in vitro methods. Antimicrobial evaluation, cytotoxic evaluation, and DNA damage checkpoints will be performed. Bovine embryos were collected from a slaughterhouse in Egypt and cultured in vitro. Sperm capacitation and acrosome reaction were assessed using sperm swim-up technique and sperm viability. The study also uses the Trypan Blue exclusion method for sperm viability and Coomassie Blue G staining for acrosome reaction. |
